目的:检测短葶山麦冬皂苷DT-13对肺癌A549细胞粘附及侵袭的作用, 及对金属基质蛋白表达的影响。方法:利用MTT法寻找到DT-13合适的作用浓度;利用A549细胞与人脐静脉内皮细胞、纤连蛋白的粘附, 评价DT-13对细胞粘附的作用;利用transwell小室,考察DT-13对细胞侵袭的作用; 采用Western Blotting方法,检测DT-13对A549细胞中金属基质蛋白-2/9的作用; 结论:DT-13在10和30 μmol·L-1时, 可显著地抑制A549细胞的粘附与侵袭,并且对A549细胞中金属基质蛋白-2和金属基质蛋白-9的表达均有抑制作用。
Objective: To detect the effect of Radix Ophiopogon japonicus Saponins (DT-13) on the adhesion and invasion of lung cancer A549 cells and its effect on the expression of matrix metalloproteinases. Methods: MTT method was used to find the appropriate concentration of DT-13. The adhesion of A549 cells to human umbilical vein endothelial cells and fibronectin was evaluated to evaluate the effect of DT-13 on cell adhesion. 13 on cell invasion; Western Blotting method was used to detect the effect of DT-13 on metal matrix protein-2/9 in A549 cells. CONCLUSION: DT-13 can be significantly inhibited at 10 and 30 μmol·L-1 A549 cell adhesion and invasion, and A549 cell matrix metalloproteinase-2 and matrix metalloproteinase-9 expression were inhibited.
