目的探讨缺氧诱导因子1α(HIF-1α)和碳酸酐酶Ⅸ(CAⅨ)表达在宫颈上皮癌变过程中的意义。方法经术后病理检查确诊宫颈病变患者89例:宫颈上皮内瘤样病变(CIN)Ⅰ-Ⅱ级28例,CINⅢ级20例,宫颈鳞癌41例;对照组为正常宫颈组织22例。应用SP免疫组化法检测HIF-1α和CAⅨ表达,分析HIF-1α和CAⅨ与宫颈癌临床病理特征间的关系。结果宫颈鳞癌、CINⅢ和CINⅠ-Ⅱ组织中的HIF-1α阳性表达率分别为75.6%、50.0%和32.1%(P<0.05),CAⅨ的阳性表达率分别为73.2%、65.0%和39.3%(P<0.05)。宫颈鳞癌组织中HIF-1α和CAⅨ的阳性表达率随临床分期的升高而增加(P<0.05);HIF-1α和CAⅨ蛋白在宫颈鳞癌中的阳性表达率呈正相关(r=0.682,P<0.05)。结论 HIF-1α和CAⅨ在CIN和宫颈癌组织中呈高表达,提示检测HIF-1α和CAⅨ对宫颈癌早期诊断有临床意义。
Objective To investigate the significance of expression of hypoxia inducible factor 1α (HIF-1α) and carbonic anhydrase Ⅸ (CAIX) in carcinogenesis of cervical epithelium. Methods Totally 89 cases of cervical lesions were diagnosed by pathological examination: 28 cases of grade Ⅰ-Ⅱ cervical intraepithelial neoplasia (CIN), 20 cases of CIN Ⅲ grade and 41 cases of squamous cell carcinoma of cervix. The control group had 22 cases of normal cervical tissue. The expression of HIF-1α and CAⅨ was detected by SP immunohistochemistry, and the relationship between HIF-1α and CAⅨ and clinicopathological features of cervical cancer was analyzed. Results The positive expression rates of HIF-1α in cervical squamous cell carcinoma, CINⅢ and CINⅠ-Ⅱ tissues were 75.6%, 50.0% and 32.1% respectively (P <0.05) .The positive expression rates of CAⅨ were 73.2%, 65.0% and 39.3% (P <0.05). The positive expression rates of HIF-1α and CAⅨ in cervical squamous cell carcinoma tissues increased with clinical stage (P <0.05). The positive expression rate of HIF-1α and CAⅨ in cervical squamous cell carcinoma tissues was positively correlated (r = 0.682, P <0.05). Conclusions HIF-1α and CAⅨ are highly expressed in CIN and cervical cancer tissues, suggesting that the detection of HIF-1α and CAⅨ is of clinical significance in the early diagnosis of cervical cancer.
