目的:为制备色泽白、黏度高的蜣螂壳聚糖,以蜣螂甲壳素为原料,探讨其脱色工艺条件。方法:首先考察脱色剂种类、H2O2浓度、pH值、固液比、反应温度等因素对壳聚糖色泽和特性黏度的影响,继而在单因素试验的基础上,采用L9(34)正交试验法优选H2O2脱色工艺的参数。结果:蜣螂甲壳素脱色的最佳工艺条件为:脱色剂H2O2浓度为15%、固液比1∶15、反应温度为45℃。结论:该工艺稳定可行,脱色效果好,可最大限度保持壳聚糖的高黏度,使其有望成为结肠定位给药的新型载体。
Objective: To prepare chitosan with high color and whiteness and high viscosity, using chitin as raw material, to explore its decolorization process conditions. Methods: Firstly, the effects of decolorant type, H2O2 concentration, pH value, solid-liquid ratio, reaction temperature and other factors on the color and intrinsic viscosity of chitosan were investigated. Then on the basis of single factor test, L9 (34) orthogonal test was used. The method optimizes the parameters of the H2O2 decolorization process. RESULTS: The optimum conditions for decolorization of chitin were: decolorizer H2O2 concentration 15%, solid-liquid ratio 1:15, reaction temperature 45 °C. Conclusion: The technology is stable and feasible, and the decolorization effect is good. It can maintain the high viscosity of chitosan to the maximum extent, making it a promising new carrier for colon positioning and administration.
