目的建立C57BL/6J小鼠肥胖模型,观察中链脂肪酸对其体脂肪的影响。方法选择4-5周龄C57BL/6J雄性小鼠40只,随机分为中链脂肪酸食用油(MCT)高脂组,中长链脂肪酸食用油(MLCT)高脂组,大豆油高脂对照组及普通饲料对照组,每组10只,喂饲15周。观察各组之间小鼠体重、饲料消耗量及食物功效比、体脂肪重和脂体比以及白色脂肪细胞数目及大小的差异,观察各组小鼠肝脏重量及脂蛋白水平的变化。结果 15周后,大豆油高脂组小鼠体重比普通饲料对照组平均增加2个标准差,且体脂肪重、脂体比均显著高于普通饲料对照组(P<0.05),造模成功。研究结束时,MLCT和MCT高脂组小鼠的体脂肪重、脂体比、肝脏重以及白色脂肪细胞数目和长径、短径均显著低于大豆油高脂对照组(P<0.05),但MLCT和MCT两组之间无显著性差异(P>0.05)。MCT组肝脏apoA1显著高于大豆油组(P<0.05),而MLCT组apoB水平显著低于大豆油组(P<0.05)。饲料消耗量及食物功效比三个高脂组之间无显著性差异(P>0.05)。结论高脂饲料可诱导C57BL/6J小鼠肥胖,含有同浓度MCFA的MLCT和MCT作用相似,可减少C57BL/6J小鼠体脂肪重、肝脏重及白色脂肪细胞体积,改善肝脏脂蛋白水平,但不影响饲料摄入量。
Objective To establish a model of obesity in C57BL / 6J mice and observe the effect of medium chain fatty acids on their body fat. Methods Forty C57BL / 6J male mice aged 4-5 weeks were randomly divided into high fat diet group (MCT), high fat diet group (MLCT), high fat diet group And normal feed control group, 10 in each group, fed for 15 weeks. The body weight, feed consumption and food efficiency, body fat weight and body fat ratio and the number and size of white adipocytes in each group were observed. The changes of liver weight and lipoprotein in the mice were observed. Results After 15 weeks, the body weight of soybean oil-fat group increased by 2 standard deviations on average compared with the control group, and body fat mass and fat body ratio were significantly higher than those of the normal control group (P <0.05) . At the end of the study, body weight, body fat ratio, liver weight, number of white adipocytes and long diameter and short diameter of mice in MLCT and MCT high fat group were significantly lower than those in high fat soybean oil control group (P <0.05) However, there was no significant difference between MLCT and MCT (P> 0.05). The apoA1 level in liver of MCT group was significantly higher than that of soybean oil group (P <0.05), while the level of apoB in MLCT group was significantly lower than that of soybean oil group (P <0.05). There was no significant difference in feed consumption and food efficiency between the three high fat diet groups (P> 0.05). Conclusion High fat diet can induce the obesity of C57BL / 6J mice. MLCT and MCT containing the same concentration of MCFA can reduce body fat mass, liver weight and white adipocyte volume, and improve hepatic lipoprotein level in C57BL / 6J mice. Does not affect feed intake.
