用HPMC为骨架材料,用湿法制粒,将易溶于水的盐酸雷尼替丁制成缓释骨架片,考察HPMC的用量、粘度、湿润剂的浓度,压片压力、片剂大小和亲水性及疏水性辅料对盐酸雷尼替丁缓释骨架片体外释药速率的影响。结果表明盐酸雷尼替丁缓释片的体外释放行为均符合Higuchi方程。HPMC用量、压片压力、片剂大小对盐酸雷尼替丁缓释片的释放速率均有显著性影响;HPMC粘度和用不同浓度乙醇制粒对盐酸雷尼替丁的释放速率影响不明显;亲水性辅料乳糖、PVP-K30和疏水性辅料EC加入均对释药速率有较大影响,前者加快药物释放而后者减慢药物释放
