目的 :观察模拟失重大鼠脑与后肢动脉血管组织血管紧张素Ⅱ 1型受体 (AT1)在基因与蛋白水平的表达是否发生差别性改变 .方法 :以 4wk尾部悬吊大鼠模型模拟失重对动脉血管的影响 .用逆转录 多聚酶链式反应 (RT PCR)检测模拟失重 (SUS)组与同步对照 (CON)组大鼠基底动脉和股动脉血管组织AT1的两种亚型 ,即AT1a和AT1b的mRNA表达水平 ;用Western印迹分析测定两种动脉组织AT1的蛋白表达水平 .结果 :SUS组大鼠基底动脉组织AT1a与AT1b的mRNA及AT1的蛋白表达水平均显著高于CON组 (P <0 0 5 ) .在股动脉组织的改变恰相反 :SUS组AT1a的mRNA表达显著低于CON组 (P <0 0 5 ) ,AT1b的表达无显著改变 ;SUS组AT1的蛋白表达也显著低于CON组 (P <0 0 5 ) .结论 :模拟失重可分别引起大鼠脑动脉与后肢动脉血管组织血管紧张素Ⅱ 1型受体的基因和蛋白表达发生增高和降低的差别性改变 .以上发现进一步支持局部肾素 血管紧张素系统在模拟失重引起的动脉血管差别性适应中可能发挥关键性调控作用
OBJECTIVE: To observe whether gene and protein expression of angiotensin Ⅱ type 1 receptor (AT1) in the brain and hind limbs of simulated weightless rats changed significantly.METHODS: A 4wk tail-suspended rat model was used to simulate the effects of weightlessness The effects of AT1a and AT1b on the basilar artery and the femoral artery tissue of rats in the simulated weightlessness (SUS) group and the synchronous control (CON) group were detected by reverse transcriptase polymerase chain reaction (RT PCR) Were detected by Western blotting.Results: The mRNA and protein expression of AT1a and AT1b in basilar artery of SUS rats were significantly higher than those in CON rats (P0.01) The changes of AT1a mRNA in SUS group were significantly lower than that in CON group (P <0.05), while the expression of AT1b was not significantly changed in SUS group. The protein expression of AT1 in SUS group was also significantly lower than that in CON group Group (P <0.05) .Conclusion: Simulated weightlessness can induce the differential expression of angiotensin Ⅱ type 1 receptor gene and protein in rat cerebral artery and hind limb arterial tissue, respectively stand by Local renin-angiotensin system may play a key regulatory role in the differential adaptation of arteries to simulated weightlessness
