目的:选用不同溶剂优化提取蓝萼香茶菜有效成分总二萜,考查蓝萼香茶菜提取物对小鼠缺氧耐受力的影响。方法:选用五种溶剂分别浸取蓝萼香茶菜地上部分,以紫外分光光度法测定各溶剂提取物总二萜含量,优化提取工艺。小鼠随机分组连续灌胃给药6天,实验组给高、中、低剂量组的蓝萼香茶菜提取物混悬液,阴性对照组给予蒸馏水,阳性对照组给予红景天。给药后进行急性脑缺血性缺氧存活时间和常压缺氧存活时间测定。结果:蓝萼香茶菜的丙酮提取物中总二萜含量相对较高;蓝萼香茶菜能够显著延长常压缺氧呼吸时间,能延长断头鼠头的张口呼吸时间,但效果不明显。结论:选取丙酮作提取溶剂的蓝萼香茶菜提取物,提取物具有提高小鼠缺氧耐受力的功效。
OBJECTIVE: To optimize the extraction of total diterpene from the active ingredients of Chayama japonicum with different solvents, and to study the effect of the extract from blue-eyed Chaikeicha on hypoxia tolerance in mice. Methods: The five solvents were used to extract the aerial parts of the blue-green tea, the total diterpene content of each solvent extract was determined by UV spectrophotometry and the extraction process was optimized. The mice were randomly divided into continuous gavage for 6 days. The experimental group was given the suspension of the extract of the calyx tea from the high, middle and low dose groups, the distilled water was given to the negative control group, and the Rhodiola was given to the positive control group. After administration, acute cerebral ischemic hypoxia survival time and hypobaric hypoxia survival time were measured. Results: The content of total diterpene in the acetone extract of the blue calyx tea dish was relatively high. The blue calyx tea cabbage could significantly prolong the time of atmospheric hypoxia respiration and prolong the oral respiration time of the caput head, but the effect was not obvious . CONCLUSION: The extract of blue-calyx Chaxingu extracted with acetone as solvent has the effect of improving hypoxia tolerance in mice.
