目的探讨Toll样受体(TLR)在人甲状腺未分化癌细胞株DRO中的表达和意义。方法应用RT-PCR的方法检测人甲状腺未分化癌细胞株DRO中TLR1-9mRNA的表达,流式细胞术分析TLR1-9蛋白表达的情况,免疫荧光共聚焦对上述结果进行再次验证。结果三种方法检测结果一致,DRO细胞表达TLR4、TLR5、TLR7和TLR9mRNA和蛋白。结论 DRO细胞表达部分TLR,其表达谱与其他肿瘤部分相同。研究DRO细胞TLR表达情况对理解其恶性行为和寻找可能存在的治疗靶点具有重要意义。
Objective To investigate the expression and significance of Toll-like receptor (TLR) in human thyroid undifferentiated carcinoma cell line DRO. Methods The expression of TLR1-9 mRNA in human thyroid undifferentiated carcinoma cell line DRO was detected by RT-PCR. The expression of TLR1-9 protein was analyzed by flow cytometry. The results were re-verified by immunofluorescence confocal analysis. Results The results of three methods were consistent. DRO cells expressed TLR4, TLR5, TLR7 and TLR9 mRNA and protein. Conclusion DRO cells express part of TLR whose expression profile is the same as that of other tumor. Studying the expression of TLR in DRO cells is of great significance for understanding its malignant behavior and finding potential therapeutic targets.
