目的:研究异种表面角膜镜片术后机体对异种角膜的免疫反应,阐明人体对异种角膜的免疫耐受机制。方法:应用流式细胞仪对9例(9只眼)接受异种表面角膜镜片术患者术后外周血T细胞的分布和自然杀伤细胞的活性进行了分析。结果:术后2个月,T细胞未被激活,NK活化程度显著提高(P<001),但未激发排斥反应。结论:猴角膜镜片虽可诱发轻微的免疫应答,但未达到增强抗原诱发的T细胞增殖应答的程度。可能与猴和人的种属相近,角膜镜片的抗原性低,抗原不易或缓慢被吞噬细胞摄取而成为致耐原,进而诱导免疫耐受有关。术后局部应用免疫抑制剂可防止移植排斥反应的发生。
OBJECTIVE: To study the immune response of xeno-cornea after heterosexual keratoplasty and to elucidate the mechanism of human immune tolerance to xeno-cornea. Methods: The distribution of T cells in peripheral blood and the activity of natural killer cells in 9 patients (9 eyes) undergoing heteroscleural keratomileusis were analyzed by flow cytometry. Results: T cells were not activated at 2 months after operation, and the activation of NK was significantly increased (P <001), but rejection was not induced. CONCLUSIONS Although monkey cornea lenses can induce a mild immune response, they do not reach the level that enhances the antigen-induced T cell proliferative response. May be similar to the monkey and human species, corneal lens antigenicity is low, the antigen is not easy or slow phagocytic uptake of protozoa, and thus induce immune tolerance. Postoperative local application of immunosuppressive agents can prevent the occurrence of transplant rejection.
