本文报道了一种测定血清生育酚水平的反相高效液相色谱法。固定相为HITACHI GEL3056(ODS,5μm);流动相为100%甲醇;流速为1.2ml/min;紫外检测波长290nm;回收率分别为93.67%(8μg/ml)和93.19%(16μg/ml);变异系数CV小于6%。并用该法测定了137名不同年龄不同性别正常人血清生育酚水平。统计学处理表明,男性和女性之间的生育酚水平无显著差异(P>0.05);男性组和女性组中,60岁以上老人与中青年相比亦无显著差异(P>0.05)。
This article reports a method for the determination of serum tocopherols by RP-HPLC. The stationary phase was HITACHI GEL 3056 (ODS, 5 μm); the mobile phase was 100% methanol; the flow rate was 1.2 ml/min; the UV detection wavelength was 290 nm; the recoveries were 93.67% (8 μg/ml) and 93.19% (16 μg/ml), respectively; The coefficient of variation CV is less than 6%. The method was used to determine the serum tocopherol levels of 137 normal people of different ages and genders. Statistical analysis showed that there was no significant difference in tocopherol levels between males and females (P>0.05). There was also no significant difference between the males and females in the elderly over the age of 60 compared with young adults (P>0.05).
