目的:建立测定降钙素基因相关肽(CGRP)的放射免疫分析法(RIA),初步应用于临床测定冠心病患者行经皮腔内冠状动脉成形术(PTCA)及植入血管内支架(IVS)后 CGRP的变化。方法:Iodogen碘化法标记Tyr~0 CGRP(24-37),选用CGRP单克隆抗体建立RIA。结果:~(125)I-Tyr~0CGRP(24-37)比活度1460Ci/mmol,放化纯度>90%,免疫活性>94%;方法灵敏度20ng/L,检测范围25-1600ng/L,平均批内、批间变异为9.2%、16.8%;测定两组术后CGRP显著升高(P<0.01),无明显峰值出现,术后24h基本降至术前水平;两组间各时间点比较无显著差异;与标准试剂盒相关性好(r=0.8962)。结论:本方法能满足临床检测要求。PTCA及植入IVS后对CGRP的影响基本相同。
OBJECTIVE: To establish a radioimmunoassay (RIA) method for the determination of calcitonin gene-related peptide (CGRP) in patients with coronary heart disease (CHD) undergoing percutaneous transluminal coronary angioplasty (PTCA) and intravascular stent implantation (IVS) Post CGRP changes. Methods: Iodogen iodide method labeled Tyr ~ 0 CGRP (24-37), the selection of CGRP monoclonal antibody RIA. Results The specific activity of ~ (125) I-Tyr ~ 0CGRP (24-37) was 1460Ci / mmol, the radiochemical purity was over 90% and the immunocompetence was over 94%. The method was sensitive to 20ng / L with the detection range of 25-1600ng / The mean intra-and inter-batch variation was 9.2% and 16.8% respectively. The postoperative CGRP levels were significantly increased in both groups (P <0.01), with no obvious peak, and basically decreased to preoperative levels at 24 hours. No significant difference compared with the standard kit (r = 0.8962). Conclusion: This method can meet the clinical testing requirements. The effects of PTCA and IVS on CGRP were essentially the same.
