从葛根提取大豆甙元,采用乙醇热回流提取的方法,醇液浓缩后用酸水解。水解液用醋酸乙醇萃取,蒸发干后,用甲醇重结晶数次,即得灰白色固体。经检测系大豆甙元(Daidzein C_(15)H_(10)O_4,7,4′二羟基异黄酮)。含有大豆甙元成分的中成药,可用薄层层析法进行鉴别试验。与标准品对照,大豆甙元斑点显色位置稳定,灵敏度高,可作为检测手段之一。
Soybean aglycone was extracted from Radix Puerariae and extracted by refluxing with ethanol. The alcohol was concentrated and then hydrolyzed with acid. The hydrolyzate was extracted with ethanol, evaporated to dryness, and recrystallized several times from methanol to give an off-white solid. It was detected to be a soybean aglycon (Daidzein C_(15)H_(10)O_4,7,4’ dihydroxyisoflavone). Chinese patent medicines containing soybean aglycon components can be identified by thin layer chromatography. Compared with the standard products, soybean aglycone spots have a stable color location and high sensitivity, which can be used as one of the detection methods.
