乙烯应答因子结合蛋白(ethylene responsive factor binding protein,ERF)属于AP2/ERF转录因子超家族中的ERF家族,是植物所特有的一类转录因子,在植物生长、发育以及应对各种环境刺激等方面均发挥重要的作用。本文从丹参中克隆得到一个丹参SmERF基因,该基因属于拟南芥和水稻ERF家族中的VII组,含有CMVII-1的蛋白模体,序列为MCGGAI(I/L),且在AP2/ERF功能域的5’端含有一个内含子。通过对丹参主产区山东、陕西、四川181个样本的检测,发现SmERF第二外显子区域存在大量的变异位点,264bp范围内共存在21个单核苷酸多态性(single nucleotide polymorphism,SNP),有14个为非同义突变,7个为同义突变。不同产地样品具有产地特异的基因型,其中有5个位点在不同产地间均表现出显著差异,聚类分析表明SmERF基因可按产地聚类。该结果显示SmERF基因有望作为丹参产地鉴别的标识基因,并为SmERF基因功能的确定奠定了基础。
Ethylene responsive factor binding protein (ERF) belongs to the ERF family in the AP2 / ERF transcription factor superfamily. Through the detection of 181 samples from the main producing areas of Danshen, Shandong, Shaanxi and Sichuan Province, there are a large number of variation sites in the second exon region of SmERF. A total of 21 single nucleotide polymorphisms , SNP), 14 were non-synonymous mutations, 7 were synonymous mutations. The samples from different areas had genotypes of origin. Among them, five loci showed significant differences among different producing areas. Cluster analysis showed that SmERF genes clustered by origin. The results showed that SmERF gene is expected to be used as a marker for the identification of Salvia miltiorrhiza and laid the foundation for the determination of SmERF gene function.
