目的探讨肺鳞癌组织中Ras相关核蛋白(Ran)的mRNA和蛋白表达水平变化及其意义。方法采用实时荧光定量PCR(RQ-PCR)和Western Blot技术检测22例不同分期及分化程度的肺鳞癌组织及其相应癌旁正常组织中Ran的表达水平,并分析其与临床病理指标的关系。结果肺鳞癌组织中Ran mRNA表达水平是癌旁正常组织的2.35倍(P<0.05),Ran在肺鳞癌组织中的蛋白表达水平高于癌旁组织。Ran在非早期(Ⅲ期)肺鳞癌组织中mRNA和蛋白的表达水平均高于早期(Ⅰ期+Ⅱ期)癌组织(P<0.05),在低分化肺鳞癌组织和高、中分化癌组织中mRNA和蛋白表达水平的差异无统计学意义(P>0.05)。结论 Ran的表达上调可能与肺癌的发生、发展相关。
Objective To investigate the changes of mRNA and protein expression of Ras-related nuclear protein (Ran) in lung squamous cell carcinoma and its significance. Methods RQ-PCR and Western Blot were used to detect the expression of Ran in 22 squamous cell carcinoma tissues with different stages and differentiations, and their corresponding normal tissues. The relationship between the expression of Ran and clinicopathological parameters was analyzed . Results Ran mRNA expression in lung squamous cell carcinoma tissues was 2.35 times that of adjacent normal tissues (P <0.05). The expression level of Ran in squamous cell carcinoma tissues was higher than that in paracancerous tissues. The expression level of Ran mRNA in non-early (stage Ⅲ) lung squamous cell carcinoma tissue was higher than that in the early stage (stage Ⅰ + Ⅱ) (P <0.05). In poorly differentiated squamous cell carcinoma and high and moderately differentiated squamous cell carcinoma, There was no significant difference in the expression of mRNA and protein in cancer tissues (P> 0.05). Conclusion The up-regulation of Ran may be related to the occurrence and development of lung cancer.
