目的:探讨中间丝蛋白nestin在视神经横断大鼠视网膜神经胶质细胞中的表达情况及可能的意义。方法:制作大鼠视神经横断模型,分别于术后1,3,7d取材,制作视网膜矢状位冰冻切片,进行GS/nestin,免疫荧光双标。提取视网膜总RNA行nestin Real-time PCR半定量分析。结果:正常大鼠视网膜中几乎看不到nestin阳性染色。术后1d,在Müller细胞上出现nestin的诱导表达,术后3d,nestin在Müller细胞上的表达进一步增强,术后1wk,nes-tin在Müller细胞上的表达保持在较高水平,Real-time PCR半定量分析与以上结果吻合。结论:视神经横断后nestin在Müller细胞上的诱导表达是Müller细胞对视网膜损伤产生的一种反应,Nestin的表达量在一定时间内与病程进展相一致。Nestin在Müller细胞上的诱导表达尤其在足板处的强烈表达可能对视网膜神经节细胞具有保护作用。
Objective: To investigate the expression of nestin, a intermediate silk protein, in retinal glia of optic nerve transected rats and its possible significance. Methods: The optic nerve transection model was made in rats. The retinal sagittal frozen sections were made at 1, 3 and 7 days postoperatively. GS / nestin and double immunofluorescence were used respectively. Retinal total RNA was extracted by semi-quantitative analysis of nestin Real-time PCR. Results: Nestin positive staining was hardly seen in normal rat retina. The expression of nestin was observed on Müller cells 1 day after transplantation. The expression of nestin on Müller cells was further enhanced 3 days after operation. The expression of nes-tin on Müller cells maintained at a high level at 1 wk postoperatively. Real-time PCR semi-quantitative analysis and the above results. CONCLUSION: The induced expression of nestin in Müller cells after optic nerve transection is a kind of response to Müller cells’ retinal damage. The expression of Nestin is consistent with the progression of the disease in a certain period of time. Inducible expression of Nestin on Müller cells, especially at the footplates, may have protective effects on retinal ganglion cells.
