目的检测信号转导与转录激活因子6(signal transducers and activators of transcription 6,STAT6)在正常食管黏膜、癌旁非典型增生和食管鳞癌组织中的表达,探讨其与食管鳞癌临床病理参数的关系。方法 2013年8月—2014年5月应用RT-PCR和Western Blotting方法检测40例食管鳞癌、40例癌旁非典型增生及20例正常食管黏膜组织中STAT6的表达。多组间比较用单因素方差分析,多样本率的比较用行列表资料的χ2检验,P<0.05为差异有统计学意义。结果 STAT6 m RNA在正常管黏膜组织、非典型增生组织和食管鳞癌组中都有表达,其相对表达强度依次升高,分别为(0.085±0.000)、(0.519±0.001)、(0.860±0.000),组间差异有统计学意义(F=2.030,P<0.05);STAT6蛋白在正常食管黏膜组织、非典型增生组织中及食管鳞癌组织中的表达量依次升高,分别为(0.339±0.001)、(0.486±0.001)、(0.624±0.001),组间差异有统计学意义(F=894.735,P<0.05);STAT6异常表达与食管癌的分级、浸润深度及淋巴结转移均密切相关(P<0.05)。结论 STAT6的表达增强可能与食管鳞癌的发生发展有关。
Objective To detect the expression of signal transducers and activators of transcription 6 (STAT6) in normal esophageal mucosa, adjacent atypical hyperplasia and esophageal squamous cell carcinoma (ESCC), and to investigate their relationship with clinicopathological parameters of esophageal squamous cell carcinoma relationship. Methods The expression of STAT6 in 40 cases of esophageal squamous cell carcinoma, 40 cases of atypical hyperplasia of carcinoma and 20 cases of normal esophageal mucosa were detected by RT-PCR and Western Blotting from August 2013 to May 2014. One-way ANOVA was used to compare the multiple groups. The multiple sample rates were compared with the chi-square test of row data. P <0.05 was considered statistically significant. Results STAT6 m RNA was expressed in normal ductal mucosa, atypical hyperplasia and esophageal squamous cell carcinoma, and the relative expression intensity of STAT6 mRNA was (0.085 ± 0.000), (0.519 ± 0.001) and (0.860 ± 0.000 ), The difference was statistically significant (F = 2.030, P <0.05). The expression of STAT6 protein in normal esophageal mucosa, atypical hyperplasia tissues and esophageal squamous carcinoma tissues were increased in turn (0.339 ± 0.309 0.001), (0.486 ± 0.001) and (0.624 ± 0.001) respectively. There was significant difference between the two groups (F = 894.735, P <0.05). The abnormal expression of STAT6 was closely related to the grade, depth of invasion and lymph node metastasis P <0.05). Conclusion The increased expression of STAT6 may be related to the occurrence and development of esophageal squamous cell carcinoma.
