目的:研究心力衰竭对心房组织瞬时受体电位通道C1(TRPC1)基因表达及其蛋白水平的影响。方法:新西兰白兔20只随机分为2组:对照组10只(其中假手术5只),心力衰竭组10只。采用主动脉瓣返流联合腹主动脉缩窄的方法制作慢性心力衰竭模型,10周后超声心动图检测心功能指标,应用RT-PCR和Western blot技术检测左心耳TRPC1基因表达和蛋白水平的变化。结果:心力衰竭组左房内径、室间隔厚度、左室后壁厚度、左室收缩和舒张末内径均明显增加(P<0.05),射血分数及左室短轴缩短率显著降低(P<0.05);与对照组相比,心力衰竭组左心耳TRPC1基因表达明显上升(P<0.05),TRPC1蛋白含量也显著高于对照组(P<0.05)。结论:兔心力衰竭后心房组织的TRPC1基因表达及其蛋白水平均明显升高,可能与心力衰竭后房性心律失常的发生有关。
Objective: To study the effect of heart failure on the gene expression and protein level of transient receptor potential channel C1 (TRPC1) in atrial tissue. Methods: Twenty New Zealand white rabbits were randomly divided into two groups: control group (n = 5) and heart failure group (n = 10). Chronic heart failure model was made by aortic regurgitation combined with abdominal aortic constriction. Ten weeks later, cardiac function was measured by echocardiography. The changes of TRPC1 gene expression and protein level in left atrial appendage were detected by RT-PCR and Western blot . Results: Left atrium diameter, interventricular septum thickness, left ventricular posterior wall thickness, left ventricular systolic and end diastolic diameters were significantly increased (P <0.05), ejection fraction and left ventricular short axis shortened significantly (P < 0.05). Compared with the control group, TRPC1 gene expression in left atrial appendage in heart failure group was significantly increased (P <0.05) and TRPC1 protein content was also significantly higher than that in control group (P <0.05). Conclusion: TRPC1 gene expression and protein level in atrial tissue after heart failure in rabbits were significantly increased, which may be related to the occurrence of atrial arrhythmia after heart failure.
