目的探讨高糖对肾小球系膜细胞细胞外基质表达的影响,以及西格列汀(SIT)对其的干预作用。方法培养肾小球系膜细胞,分为对照(Con)组、高糖(HG)组,以及高糖联合不同浓度的SIT组(HG+S1组、HG+S5组、HG+S10组和HG+S20组)。培养24 h、48 h后,用MTT法测定细胞增殖程度,ELISA检测细胞培养上清层粘连蛋白(LN)、大鼠Ⅳ型胶原蛋白(Col-Ⅳ)、转化生长因子β1(TGF-β1)蛋白表达的变化。结果高糖培养24 h、48 h后,可观察到不同浓度的SIT均可抑制高糖诱导的系膜细胞增殖;SIT组与HG组比较,差异有统计学意义;Con组肾小球系膜细胞可分泌一定量的LN、Col-Ⅳ和TGF-β1,高糖刺激后LN、Col-Ⅳ和TGF-β1分泌增加,而SIT对系膜细胞分泌物的抑制随其浓度的升高而增强。结论SIT可抑制高糖条件下系膜细胞的增殖,且能降低高糖条件下LN、Col-Ⅳ、TGF-β1的表达,SIT可能对糖尿病患者的肾脏有一定的保护作用。
Objective To investigate the effect of high glucose on the expression of extracellular matrix in glomerular mesangial cells and the effect of sitagliptin (SIT) on it. Methods Glomerular mesangial cells were cultured and divided into control group (Con group), high glucose (HG) group and high glucose group with different concentrations of SIT group (HG + S1 group, HG + S5 group, HG + S10 group and HG + S20 group). After cultured for 24 h and 48 h, the degree of cell proliferation was determined by MTT assay. Results After cultured with high glucose for 24 h and 48 h, different concentrations of SIT could inhibit the proliferation of mesangial cells induced by high glucose. The difference was statistically significant between SIT group and HG group. In Con group, The cells could secrete a certain amount of LN, Col-Ⅳ and TGF-β1. After high glucose stimulation, the secretion of LN, Col-Ⅳ and TGF-β1 increased, while the inhibitory effect of SIT on the secretion of mesangial cells increased with the concentration of LN . Conclusion SIT can inhibit the proliferation of mesangial cells under high glucose and reduce the expression of LN, Col-Ⅳ and TGF-β1 under high glucose. SIT may have a protective effect on the kidney of diabetic patients.
