水稻花药培养是诱导单倍体的一种十分有效的方法。中国运用花药培养已育成20多个水稻新品种。目前,这种技术虽已达到一定的水平,但仍有一些问题尚待解决。其中最严重的问题之一是再生过程中产生白化苗,从而明显地降低了单倍体育种的效率。新关(1977)和大村(1980)报导了对氟苯基丙氨酸(PFP)可减少野生稻、草莓和葡萄的染色体数。本试验检查了PFP对水稻栽培品种“日本晴”染色体数减少的影响。置无菌糙米于N6琼脂培养基上,N6琼脂培养基含20微克分子浓度的2,4-D和五种浓度(0、25、50、250、和500微克分子)的PFP。
Rice anther culture is a very effective method to induce haploid. China has used anther culture to breed more than 20 new rice varieties. At present, although this technology has reached a certain level, there are still some problems to be solved. One of the most serious problems is the generation of albino seedlings during regeneration, thus significantly reducing the efficiency of haploid breeding. Shin-Kuan (1977) and Omura (1980) reported that p-fluorophenylalanine (PFP) reduced chromosome numbers in wild rice, strawberries and grapes. This experiment examined the effect of PFP on the reduction of chromosome number in “Nipponbare” rice cultivars. Sterile brown rice was placed on N6 agar medium containing 20 micromolar concentrations of 2,4-D and five concentrations (0, 25, 50, 250, and 500 micromolar) of PFP.
