目的 定量检测早期乳腺癌患者外周血的肿瘤细胞含量 (PBCC )。方法 采用人细胞角蛋白抗体和CD 45抗体双色免疫标记外周血标本 ,应用多参数流式细胞仪方法检测早期乳腺癌患者手术前后外周血中肿瘤细胞含量。结果 术前PBCC与肿瘤大小、腋窝淋巴结状况、病理分期及脉管瘤栓均具有明显相关性 (P <0 .0 1) ,而术后PBCC与腋窝淋巴结状况、病理分期和脉管瘤栓具有相关性 (P <0 .0 5 ) ,而与肿瘤大小无关。结论 采用流式细胞术检测肿瘤患者外周血中肿瘤细胞具有高灵敏度、定量等优势 ,为肿瘤治疗提供了有效的连续监测手段 ,对乳腺癌的治疗和预后判断具有重要应用价值。
Objective To quantitatively detect the tumor cell content (PBCC) of peripheral blood in patients with early breast cancer. Methods Peripheral blood samples were immunolabeled with human cytokeratin antibody and CD45 antibody. Multi-parameter flow cytometry was used to detect the content of tumor cells in peripheral blood of patients with early breast cancer. Results Preoperative PBCC was significantly correlated with tumor size, axillary lymph node status, pathological stage, and vascular tumor embolus (P < 0.01). Postoperative PBCC and axillary lymph node status, pathological staging, and vascular tumor emboli had Correlation (P < 0.05), regardless of tumor size. Conclusion The use of flow cytometry to detect tumor cells in peripheral blood of patients with tumor has high sensitivity and quantitative advantages, which provides an effective continuous monitoring method for cancer treatment, and has important application value for the treatment and prognosis of breast cancer.
