本文用细胞培养方法,观察脑血管内皮细胞粘附分子(ICAM-1)的表达和内皮细胞与血液单核细胞的粘附作用。结果显示,10u/mlTNF-α即可使内皮细胞ICAM-1表达增加,自发性高血压大鼠(SHR)增加的程度大于正常血压大鼠(WKY)。干扰素(IFN)和白细胞介素1(IL-1)也可使ICAM-1表达增加。高浓度细胞因子均使SHR和WKY内皮细胞ICAM-1表达增加,但二者之间无明显差别。经LPS或IL-1和TNF-α刺激的SHR内皮细胞与单核细胞的粘附率明显高于WKY。结果证实,TNF-α,IL-1和IFN可使SHR和WKY脑血管内皮细胞ICAM-1表达和单核细胞粘附增加,两组动物细胞相比,SHR内皮细胞ICAM-1表达及粘附率增加较多,提示SHR脑血管内皮细胞可能对细胞因子的敏感性升高。
In this paper, the cell culture method was used to observe the expression of ICAM-1 and the adhesion of endothelial cells to blood mononuclear cells. The results showed that 10u / ml TNF-αcan increase the expression of ICAM-1 in endothelial cells, and the degree of spontaneous hypertensive rats (SHR) increased more than normal blood pressure rats (WKY). Interferon (IFN) and interleukin 1 (IL-1) also increase ICAM-1 expression. The results confirmed that TNF-α, IL-1 and IFN could increase ICAM-1 expression and monocyte adhesion in SHR and WKY cerebrovascular endothelial cells.
