目的:研究补中益气汤对创伤大鼠应激反应的影响,初步探索其作用机制。方法:将30只SD大鼠随机分成3组,每组均为10只,即分成Ⅰ组正常对照组、Ⅱ组模型对照组、Ⅲ组中药治疗组。模型对照组和中药治疗组制成骨折创伤模型,伤后1h开始给Ⅰ组、Ⅱ组经胃灌注生理盐水,给Ⅲ组经胃灌注中药,每次均为0.5mL,每日2次。于创伤后6h、1、3、5、7天采静脉血测定应激激素COR、NE和细胞因子TNF-α、IL-4的水平。结果:Ⅱ组COR在各时间点较Ⅰ组显著升高(P<0.05或P<0.01),上升缓慢而持久;Ⅱ组NE在伤后6h、1天较Ⅰ组显著升高(P<0.01),整个时间点NE上升快,但下降也快;Ⅱ组TNF-α在伤后显著升高(P<0.01),在整个时间点上无显著变化;Ⅱ组细胞因子IL-4在创伤后明显降低(P<0.01),且回升缓慢。Ⅲ组对血中应激激素COR在伤后3、5、7天的升高有显著控制作用,而NE在伤后6h、1天的升高有显著控制作用,与Ⅱ组比较差异有显著性(P<0.05或P<0.01)。Ⅲ组对血中细胞因子TNF-α的升高和对细胞因子IL-4的降低均有显著控制作用,与Ⅱ组比较差异均有显著性(P<0.05或P<0.01)。结论:补中益气汤能有效调理创伤后应激反应的发生和发展。
Objective: To study the effect of Buzhong Yiqi Decoction on the stress response in traumatic rats and to explore its mechanism. Methods: Thirty SD rats were randomly divided into three groups, each group was 10, which divided into Ⅰ normal control group, Ⅱ model control group, Ⅲ Chinese medicine treatment group. The model control group and the Chinese medicine treatment group were made into the model of fracture trauma. The rats in group Ⅰ and group Ⅱ were given gastric perfusion with saline at 1h after injury, and the rats in group Ⅲ were treated with stomach perfusion with 0.5mL twice daily. The levels of COR, NE and cytokines TNF-α and IL-4 were measured at 6 h, 1, 3, 5 and 7 days after trauma. Results: The COR in group Ⅱ was significantly higher than that in group Ⅰ (P <0.05 or P <0.01) at each time point, and the increase was slow and persistent. The NE of group Ⅱ was significantly higher than that of group Ⅰ at 6 and 1 days after injury (P <0.01) ), The whole time point NE increased rapidly, but the decline was also fast; TNF-α in group Ⅱ was significantly increased after injury (P <0.01), no significant changes in the whole time point; Ⅱ group of cytokines IL-4 in trauma Decreased significantly (P <0.01), and recovered slowly. Group Ⅲ had a significant effect on the increase of blood pressure hormone COR at days 3, 5, and 7 after injury, while NE had a significant control effect at 6 and 1 day after injury, which was significantly different from that of group Ⅱ (P <0.05 or P <0.01). The level of TNF-α in blood and the decrease of cytokine IL-4 in group Ⅲ were significantly lower than those in group Ⅲ (P <0.05 or P <0.01). Conclusion: Buzhong Yiqi Decoction can effectively regulate the occurrence and development of post-traumatic stress response.
