目的研究利培酮的经皮渗透性并优化其贴剂处方。方法比较几种促渗剂的促渗作用,筛选出良好的透皮促渗剂,通过正交设计得到最优的处方组成,并评价贴剂的性能。结果 10%油酸相对于其他促渗剂有更好的促渗效果,最佳处方中癸二酸二丁酯的含量为25%、丁二酸为6%、载药量为6%,所制备的利培酮透皮贴剂的体外透皮速率为28.252μg·cm-2.h-1,且各项性能良好。结论所得处方中各因素的组合有利于利培酮的经皮吸收。
Objective To study the transdermal permeability of risperidone and to optimize its patch formulation. Methods To compare the promoting effects of several penetration enhancers, select a good transdermal penetration enhancer, obtain the optimal prescription composition by orthogonal design, and evaluate the performance of the patch. Results 10% oleic acid had a better effect of promoting penetration compared with other penetration enhancers. The best prescription was dibutyl sebacate 25%, succinic acid 6% and drug loading 6% The transdermal delivery rate of Risperidone transdermal patch prepared in the experiment was 28.252μg · cm-2.h-1, and all the properties were good. Conclusion The combination of various factors in the prescription is beneficial to the transdermal absorption of risperidone.
