目的探讨胶质瘤原代培养细胞中基质金属蛋白活性酶-2(matrix metalloproteinase-2,MMP-2)对肿瘤细胞侵袭力的影响。方法对21例Ⅳ级胶质瘤组织进行原代培养并传代,通过明胶酶谱检测MMP-2酶原及活性酶含量,Boyden小室侵袭实验检测细胞侵袭力,分析MMP-2酶原及活性酶与肿瘤细胞侵袭能力之间的关系。结果 MMP-2活性酶的含量与肿瘤细胞侵袭力正相关(r>0,P<0.05),pro-MMP-2含量与肿瘤细胞侵袭力的相关性不具有统计学意义(P>0.05)。结论Ⅳ级胶质瘤中MMP-2活性酶是决定肿瘤细胞侵袭力的关键因素。
Objective To investigate the effect of matrix metalloproteinase-2 (MMP-2) on invasiveness of tumor cells in primary cultured glioma cells. Methods Twenty-one grade IV glioma tissues were primary cultured and passaged. The content of MMP-2 and the activity of MMP-2 were detected by gelatin zymography. The cell invasiveness was assayed by Boyden chamber invasion assay. The activity of MMP-2 and active enzyme And tumor cell invasion ability. Results The level of MMP-2 activity was positively correlated with invasiveness of tumor cells (r> 0, P <0.05). The correlation between pro-MMP-2 level and invasiveness of tumor cells was not statistically significant (P> 0.05). Conclusion The activity of MMP-2 in grade IV glioma is a key factor in determining invasiveness of tumor cells.
