目的确定ESM-1mRNA在肾癌中的定位,探讨其表达在肾癌生物学特性中的意义。方法采用地高辛标记的原位杂交技术检测ESM-1mRNA在50例肾癌、30例癌旁正常肾组织中的表达状况,并分析其与临床病理特征的关系。结果ESM-1mRNA主要定位于血管内皮细胞的胞质伴胞核中,在正常肾组织与肾癌中均有表达,部分表达在肾小管上皮和肾癌细胞。肾癌间质血管内皮的ESM-1mRNA高表达率(52.00%)显著高于正常肾组织的血管内皮(13.33%);肾透明细胞癌与颗粒细胞癌中ESM-1mRNA的表达差异有显著性,而与患者的性别、年龄、肿瘤的组织学分级、淋巴结转移和肿块大小无明显相关。结论ESM-1mRNA表达可作为新生血管内皮的标志物,可能与肾癌的血管生成及发生、发展有关。
Objective To determine the localization of ESM-1 mRNA in renal cell carcinoma and to explore its significance in the biological characteristics of renal cell carcinoma. Methods The expression of ESM-1 mRNA in 50 cases of renal cell carcinoma and 30 cases of adjacent normal renal tissues was detected by digoxigenin-labeled in situ hybridization. The relationship between ESM-1 mRNA and the clinicopathological features was analyzed. Results ESM-1 mRNA mainly located in the cytoplasm of vascular endothelial cells with normal renal tissue and renal cell carcinoma were expressed, and partially expressed in renal tubular epithelial cells and renal cell carcinoma. The high expression rate of ESM-1 mRNA in renal interstitial vascular endothelium (52.00%) was significantly higher than that in normal renal tissues (13.33%). The difference of ESM-1 mRNA expression in renal clear cell carcinoma and granulosa cell carcinoma was significant However, there was no significant correlation with the patient’s gender, age, histological grade, lymph node metastasis and tumor size. Conclusion The expression of ESM-1 mRNA can be used as a marker of neovascular endothelial cells and may be related to angiogenesis, development and progression of renal cell carcinoma.
