目的采用HPLC法测定附片中总酯型生物碱的含量。方法采用Phenomenex Luna C18色谱柱(150mm×4.6mm,5μm),流动相为甲醇-0.05moL·L-1磷酸二氢钾-醋酸-异丙醇(67∶173∶4∶4),流速1.0mL·min-1,检测波长230nm,柱温35℃。结果苯甲酸0.016~0.080μg与峰面积的线性关系良好,平均回收率为97.6%,RSD=0.9%(n=5)。结论所建方法操作简便,结果可靠,可为附片中总酯型生物碱的含量测定提供参考方法。
OBJECTIVE To determine the content of total ester alkaloids in epimedium tablets by HPLC. Methods Phenomenex Luna C18 column (150mm × 4.6mm, 5μm) was used. The mobile phase was methanol-0.05moL·L -1 potassium dihydrogen phosphate-acetic acid-isopropanol (67:173:4:4) Min-1, detection wavelength 230nm, column temperature 35 ℃. Results The linear range of 0.016 ~ 0.080μg benzoic acid to peak area was good with an average recovery of 97.6% and a RSD of 0.9% (n = 5). Conclusion The method is simple, reliable and can provide a reference method for the determination of total ester alkaloids in the pellets.
