目的比较高效液相色谱法(HPLC)和超高效液相色谱-串联质谱法(UPLC-MS/MS)2种方法测定玉米油中玉米赤酶烯酮的优点、缺点。方法分别以HPLC和UPLC-MS/MS进行测定,考察2种方法测定结果的相关性,比较2种方法的性能指标。结果 2种方法的定量结果差异无统计学意义(P>0.05),具有良好的相关性;采用高效液相色谱法时,对于检出限≥5.0μg/kg的样品,2种方法的检出率基本相符;对于检出限<5.0μg/kg的样品,由于UPLC-MS/MS具有更高的灵敏度,该方法的检出率高于HPLC。结论 2种方法均能应用于玉米油中玉米赤酶烯酮的测定。高效液相色谱法因具有准确、实用和配置成本相对低的优点,更适合推广,可用于玉米油中玉米赤酶烯酮的常规监测;超高效液相色谱-串联质谱法的灵敏度高,但配置成本高,更适用于确认验证。
OBJECTIVE To compare the advantages and disadvantages of two methods for the determination of zearalenone in corn oil by high performance liquid chromatography (HPLC) and ultra performance liquid chromatography-tandem mass spectrometry (UPLC-MS / MS). Methods HPLC and UPLC-MS / MS were used to determine the correlation between the two methods. The performance of the two methods was compared. Results There was no significant difference in the quantitative results between the two methods (P> 0.05), and there was a good correlation between the two methods. When the HPLC method was used, for the samples with the detection limit of ≥ 5.0 μg / kg, the two methods were detected The results showed that UPLC-MS / MS was more sensitive than HPLC for the samples with the detection limit of <5.0μg / kg. Conclusion Both methods can be applied to the determination of maize erythroketene in corn oil. High performance liquid chromatography (HPLC) is more suitable for generalization due to its advantages of accuracy, practicality and relatively low configuration cost. It can be used for the routine monitoring of maize erythronenone in corn oil. Ultra-performance liquid chromatography-tandem mass spectrometry has high sensitivity but High configuration costs, more suitable for validation verification.
