目的:检测YY1在胃癌组织中的表达情况,探讨YY1对人胃癌细胞株SGC-7901增殖、凋亡的影响以及可能的机制。方法:利用定量PCR和免疫组织化学分别检测标本中YY1的mRNA和蛋白表达水平。利用慢病毒表达载体系统在胃癌细胞株SGC-7901中上调YY1,运用CCK-8、流式细胞术、平板克隆实验检测YY1对细胞的增殖、凋亡以及克隆形成能力的影响。结果:胃癌组织中YY1的mRNA和蛋白表达水平明显低于癌旁组织。与空载对照组(SGC-7901-Empty Vector,SGC-7901-EV)和与正常对照组(SGC-7901)相比,过表达YY1组细胞(SGC-7901-YY1)的增殖和平板克隆形成能力明显减弱,细胞凋亡增加。结论:YY1与胃癌的发生发展有一定的关系,可能起到抑癌基因的作用,YY1基因可能成为胃癌靶向治疗的新的潜在靶点。
OBJECTIVE: To detect the expression of YY1 in gastric cancer and investigate the effect of YY1 on the proliferation and apoptosis of human gastric cancer cell line SGC-7901 and its possible mechanism. Methods: Quantitative PCR and immunohistochemistry were used to detect the expression of YY1 mRNA and protein in the specimens. YY1 was upregulated in gastric cancer cell line SGC-7901 by lentivirus expression vector system. The effects of YY1 on proliferation, apoptosis and clonality were detected by CCK-8, flow cytometry and plate clone assay. Results: The mRNA and protein expression of YY1 in gastric cancer tissues was significantly lower than that in paracancerous tissues. Proliferation and plate clonality of the over-expressed YY1 cells (SGC-7901-YY1) compared to the SGC-7901-empty vector (SGC-7901-EV) and the normal control (SGC-7901) Ability was significantly weakened, increased apoptosis. Conclusion: YY1 has a certain relationship with the occurrence and development of gastric cancer, may play a role in tumor suppressor gene, YY1 gene may be a new potential target for targeted therapy of gastric cancer.
