目的探讨不同剂量氯化甲基汞蓄积对成年大鼠脑组织胶质纤维酸性蛋白(glial fibrillary acidic protein,GFAP)、髓鞘碱性蛋白(myelin basic protein,MBP)的影响。方法 120只SD大鼠随机分为正常对照组、氯化甲基汞小剂量组(0.5mg/kg)、氯化甲基汞中剂量组(1.0mg/kg)、氯化甲基汞大剂量组(2.0mg/kg)。每天灌胃1次,连续染毒,各组染毒时间分别为1、2、4周。采用免疫组织化学方法观察不同时间点各组大鼠脑组织中GFAP、MBP的变化。结果各染毒时间组中氯化甲基汞剂量增加,大鼠脑组织GFAP表达升高和MBP表达降低,差异均有统计学意义(P<0.01)。同一剂量组不同染毒时间比较,随染毒时间延长,GFAP表达升高;MBP表达降低,差异均有统计学意义(P<0.01)。结论氯化甲基汞促进星形胶质细胞增殖,破坏髓鞘,并与汞的蓄积量与染毒时间相关。
Objective To investigate the effects of different concentrations of methylmercury chloride on glial fibrillary acidic protein (GFAP) and myelin basic protein (MBP) in adult rat brain. Methods One hundred and twenty SD rats were randomly divided into normal control group, methylmercury chloride low dose group (0.5mg / kg), methylmercury methylmercury dose group (1.0mg / kg), methylmercury methyl chloride high dose Group (2.0 mg / kg). Gavage 1 times a day, continuous exposure, each group exposure time were 1,2,4 weeks. Immunohistochemistry was used to observe the changes of GFAP and MBP in brain tissue of rats in different time points. Results The dose of methylmercury chloride increased in each exposure time group, while the expression of GFAP and the expression of MBP decreased in rat brain tissue (P <0.01). Compared with the same dosage group, GFAP expression increased and MBP expression decreased with the prolongation of exposure time, the difference was statistically significant (P <0.01). Conclusion Methylmercury chloride can promote astrocyte proliferation and destroy myelin sheath, and is related to the accumulation of mercury and exposure time.
