[目的]探讨望春花挥发油(the Volatile Oil of Magnolia biondii pamp.)对BALB/c小鼠过敏性哮喘气道炎症的作用。[方法]健康BALB/c小鼠50只,雌雄兼用,体重(20±2)g,按照体重、性别分层随机分为正常对照(A)组、模型对照(B)组、阳性对照(C)组、望春花挥发油大剂量(D)组和望春花挥发油小剂量(E)组。A组以生理盐水代替抗原进行致敏,B、C、D、E组每鼠于第1、14、28d分3次腹腔注射(ip)含卵白蛋白(OVA)和福氏完全佐剂的生理盐水溶液致敏,1次/d、持续14d雾化OVA激发,建立哮喘气道炎症模型。首次激发当日开始灌胃给药(ig),1次/d,连续14d。于末次激发24h后处置动物,采制标本与指标检测,观察望春花挥发油对其支气管肺泡灌洗液(BALF)中白细胞(WBC)数与分类计数、蛋白含量、肺组织病理改变及肺细支气管黏膜浸润的嗜酸性粒细胞(Eos)数的影响。[结果](1)D组小鼠肺BALF中Eos比值为(3.17±1.33)%、蛋白含量为(0.34±0.06)×109g/L,分别与B组的Eos比值和蛋白含量[(4.17±1.47)%、(0.58±0.15)×109g/L]比较,差异均有统计学意义(P均﹤0.05);D组小鼠肺细支气管黏膜组织中Eos数(个/视野)为(8.50±1.87),与B组(12.16±2.14)比较,差异均有统计学意义(P﹤0.05);E组动物Lym%呈下降趋势,但与B组比较差异无统计学意义(P﹥0.05),是否具有免疫抑制效应,尚待进一步研究;E组小鼠其余指标与B组比较差异无统计学意义(P均﹥0.05)。(2)镜下观,与B组比较,D组小鼠细支气管上皮和基底膜变性明显减轻,固有层内小静脉、毛细血管无明显扩张,黏膜增厚不明显,细支气管黏膜和管腔内的炎细胞明显减少。[结论]望春花挥发油可明显抑制实验性BALB/c小鼠哮喘气道浸润Eos细胞数,减轻哮喘气道的炎症反应。
[Objective] To investigate the effect of the Volatile Oil of Magnolia biondii pamp. On allergic asthma airway inflammation in BALB / c mice. [Results] (1) The Eos ratio in BALF was (3.17 ± 1.33)% and the protein content was (0.34 ± 0.06) × 109g / L in group D, 1.47%, (0.58 ± 0.15) × 109g / L], the differences were statistically significant (P <0.05). The number of Eos in lung tissue of mice in group D was (8.50 ± 1.87). Compared with group B (12.16 ± 2.14), the difference was statistically significant (P <0.05). (2) Compared with group B, the degeneration of the bronchiolar epithelium and basement membrane in group D was significantly reduced. There was no obvious dilatation of small veins and capillaries in the lamina propria, and mucosal thickening was not obvious. The bronchiolar mucosa and lumen Within the inflammatory cells decreased significantly. [Conclusion] The Volatile Oil of Vetch could obviously inhibit the number of Eos cells infiltrated by airway in experimental BALB / c mice and relieve the airway inflammation of asthma.
