背景apoE基因敲除或apoE2转基因小鼠是研究高三酰甘油血症与动脉粥样硬化(atherosclerosisAS)较好模型,但缺乏方便实用的动物模型。目的探讨链脲佐菌素(streptozotocin,STZ)和饮食结构对新西兰大白兔血脂组分及AS的作用,以寻找方便实用的研究高三酰甘油血症与AS动物模型。设计随机对照的实验研究。地点、材料和干预实验在解放军第三军医大学西南医院中心实验室完成。实验选用健康雄性新西兰大白兔。采用随机抽签法分为4组标准兔小颗粒饲养(SC)组;高胆固醇饲料饲养(HC)组;STZ处理后饲养标准兔小颗粒(STZ+SC)组;STZ处理后饲养高胆固醇饲料(STZ+HC)组。比较STZ和高胆固醇饲料对新西大白兔血脂组分和AS病变的作用,并分析AS病变局部清道夫受体A(scavengerreceptor,SRA)表达与泡沫细胞分布的关系。主要观察指标观察各组大白兔血浆脂蛋白和载脂蛋白水平和主动脉内膜病理变化,并分析血管内膜粥样硬化斑块局部SRA表达与泡沫细胞分布的关系。结果STZ组大白兔血浆三酰甘油[(2.59±0.64)mmol/L]较SC组[(1.12±0.34)mmol/L]轻-中度升高(P<0.05)、HDL-C降低及大动脉AS病变;STZ+HC组大白兔的血浆总胆固醇水平[(14.18±2.72)mmol/L]与HC组[(13.74±1.43)mmol/L]相似,而前者的血浆三酰甘油水平[(3.84±0.73)mmol/L]显著高
Background apoE knockout or apoE2 transgenic mice are a good model for the study of hypertriglyceridemia and atherosclerosisAS, but lack of a convenient and practical animal model. Objective To investigate the effect of streptozotocin (STZ) and dietary structure on lipid composition and AS in New Zealand white rabbits in order to find a convenient and practical method for the study of hypertriglyceridemia and AS animal model. Design of Randomized Controlled Experimental Study. Location, materials and intervention experiments were performed at the Central Laboratory of Southwest Hospital of the Third Military Medical University. Experimental selection of healthy male New Zealand white rabbits. The rabbits were divided into 4 groups: standard small rabbit pellet (SC) group, high cholesterol feed (HC) group, STZ + STZ group, STZ group STZ + HC) group. The effects of STZ and high-cholesterol diet on lipid composition and AS pathological changes of New Zealand white rabbits were compared. The relationship between scavenger receptor A (SRA) expression and foam cell distribution was analyzed. MAIN OUTCOME MEASURES Plasma lipoprotein and apolipoprotein levels and pathological changes of aortic intima were observed in rabbits in each group. The relationship between local SRA expression and foam cell distribution was analyzed. Results Compared with SC group [(1.12 ± 0.34) mmol / L], triglyceride (2.59 ± 0.64 mmol / L) in STZ group was mild to moderate (P <0.05) and HDL- AS lesions in rabbits in STZ + HC group were significantly higher than that in HC group [(14.18 ± 2.72) mmol / L] [(13.74 ± 1.43) mmol / L], while the former plasma triglyceride levels ± 0.73) mmol / L] was significantly higher
