【目的】探讨DJ-1基因在喉鳞状细胞癌组织及Hep-2细胞系中的表达特征。【方法】采用间接免疫荧光方法,检测15例喉鳞状细胞癌手术切除癌组织、癌旁组织、正常黏膜标本和Hep-2细胞系的DJ-1蛋白。【结果】喉鳞状细胞癌组织与Hep-2细胞系中DJ-1蛋白均有表达;DJ-1蛋白在喉鳞状细胞癌组织的表达水平分别高于癌旁组织、正常黏膜组织,差异有统计学意义(F=8.098,P=0.002);低分化喉癌较高分化喉癌的DJ-1水平增高,差异有统计学意义(t=3.099,P=0.008);喉癌的临床分期和年龄与DJ-1蛋白的表达没有相关性(t=0.476,P=0.642;t=0.019,P=0.985)。【结论】喉鳞状细胞癌组织及Hep-2细胞系中均有DJ-1蛋白的表达,喉鳞癌组织内的表达高于癌旁组织和黏膜组织;DJ-1基因在喉鳞状细胞癌组织中的表达可能提示它参与了喉癌的发生发展的有关过程,为基因治疗提供了理论基础。
【Objective】 To investigate the expression of DJ-1 gene in laryngeal squamous cell carcinoma and Hep-2 cell lines. 【Methods】 DJ-1 protein was detected by indirect immunofluorescence in 15 specimens of laryngeal squamous cell carcinoma, resected from cancer tissue, adjacent normal tissue, normal mucosa and Hep-2 cell line. 【Results】 The expression of DJ-1 protein in laryngeal squamous cell carcinoma and Hep-2 cell lines was both higher than that in normal laryngeal squamous cell carcinoma The difference was statistically significant (t = 3.098, P = 0.008). The clinical stage of laryngeal cancer was statistically significant (F = 8.098, P = 0.002) And age did not correlate with DJ-1 protein expression (t = 0.476, P = 0.642; t = 0.019, P = 0.985). 【Conclusions】 The expression of DJ-1 protein in both laryngeal squamous cell carcinoma and Hep-2 cell lines was higher than that in adjacent tissues and mucosa.
