以 N-羟基琥珀酰亚胺 - α-萘乙酸酯 (SINA)为氨基酸的柱前衍生试剂 ,反相高效液相色谱分离测定了 1 5种氨基酸。采用含 1 0 mmol/ L p H 5.0的乙酸 -乙酸钠缓冲溶液的甲醇 -乙酸乙酯 -水 (1 0 / 2 / 88,V/ V/ V)溶液为流动相体系 ,分离测定了 7种氨基酸 ;用甲醇 -乙酸乙酯 -水 (2 6/ 2 / 77,V/ V/ V)分离测定了 5种氨基酸 ;用甲醇 -乙酸乙酯 -水 (45/ 2 / 53,V/ V/ V)分离测定了 3种氨基酸。检测限均达到 pmol级 ,且重现性好
Fifteen kinds of amino acids were separated and determined by reversed-phase high-performance liquid chromatography with N-hydroxysuccinimide-α-naphthalene acetate (SINA) as the pre-column derivatization reagent. The mobile phase consisted of methanol-ethyl acetate-water (10/88/8, V / V / V) solution containing 10 mmol / L p H 5.0 in acetic acid-sodium acetate buffer solution. Seven Five amino acids were determined by methanol-ethyl acetate-water (26/2/77, V / V / V) V) Three amino acids were isolated and determined. Detection limit reached pmol level, and good reproducibility
