目的 探讨miR-101对卵巢癌SKOV3细胞增殖及侵袭能力的影响.方法 将卵巢癌SKOV3细胞分为miR-101抑制剂组、miR-101模拟物组和对照组,分别将miR-101抑制剂或miR-101模拟物转染至miR-101抑制剂组或miR-101模拟物组细胞,对照组细胞转染无关序列,Real-time PCR法检测各组细胞miR-101表达水平,分别用MTT方法与Transwell细胞侵袭实验检测转染后各组SKOV3细胞的增殖与侵袭能力变化.Real-time PCR与Western blot实验分别检测各组细胞c-Fos mRNA和蛋白表达水平.结果 转染miR-101模拟物后,SKOV3细胞miR-101的表达水平显著升高,SKOV3细胞的增殖与侵袭能力显著降低,c-Fos mRNA及蛋白的表达水平显著降低(P<0.01);转染miR-101抑制剂后,SKOV3细胞miR-101的表达水平显著降低,SKOV3细胞的增殖与侵袭能力显著升高,c-Fos mRNA及蛋白的表达水平显著升高(P<0.01).结论 miR-101抑制卵巢癌SKOV3细胞的增殖和侵袭可能与下调c-Fos的表达相关.“,”Objective To investigate the effect of miR-101 on the proliferation and invasion of ovarian cancer SKOV3 cells.Methods The ovarian cancer SKOV3 cells were divided into miR-101 inhibitor group,miR-101 mimics group and control group,miR-101 inhibitors or miR-l01 mimics were transfected into SKOV3 cells in miR-101 inhibitor group or miR-101 mimics group respectively,control group was transfected with unrelated sequence.The miR-101 level was detected by real-time PCR,proliferation and invasion ability of SKOV3 cells were detected by MTT method and transwell cell invasion assay.Real-time PCR and Western blot were used to detect the expressions of c-Fos mRNA and protein respectively.Results The expression level of miR-l01 in SKOV3 cells was increased,the proliferation and invasion capacity of SKOV3 cells,the expression level of c-fos mRNA and protein were significantly decreased (P<0.01)in mir-101 mimics than in control.The expression level of miR-101 in SKOV3 cells was decreased,the proliferation and invasion capacity of SKOV3 cells,the expression level of c-fos mRNA and protein was significantly increased (P<0.01) in miR-101 inhibitors than in control.Conclusion The inhibition of miR-101 on the proliferation and invasion of ovarian cancer SKOV3 cells might be related to down-regulating the expression of c-Fos.
