目的研究硼卡钠(BSH)对辐射损伤小鼠的抗氧化作用,探讨硼卡钠辐射防护作用的抗氧化机制。方法将BALB/c小鼠按体重随机分为正常对照组、照射对照组、阳性药组(WR2721,200 mg/kg)、BSH低剂量组(20mg/kg)、BSH中剂量组(40 mg/kg)、BSH高剂量组(80 mg/kg),每组7只。各组动物腹腔注射,给药体积均为0.2 ml/20g;正常对照组和照射对照组给予生理盐水;阳性药组照射前连续给药2d,每天1次,其余各组照射前24h给药1次。用60Coγ射线对小鼠进行一次性全身照射,照射剂量为6 Gy,剂量率为0.8 Gy/min。照射后第14天采集小鼠外周血并分离血清,分别检测血清SOD活性和MDA含量。结果 BSH低、中、高剂量组小鼠外周血血清SOD活性均显著高于照射对照组(P<0.05);血清MDA值,低剂量组显著低于照射对照组(P<0.05),中、高剂量组非常显著低于照射对照组(P<0.01)。结论 BSH具有显著的抗氧化作用,提示BSH的抗氧化能力是其抗辐射作用的机理之一。
Objective To study the anti-oxidative effect of borocarp sodium (BSH) on radiation-injured mice and to explore the anti-oxidative mechanism of the radioprotective effect of boron sodium. Methods BALB / c mice were randomly divided into normal control group, irradiation control group, positive group (WR2721, 200 mg / kg), BSH low dose group (20 mg / kg), high-dose BSH group (80 mg / kg), 7 in each group. The animals in each group were injected intraperitoneally with the volume of 0.2 ml / 20 g. The normal control group and the irradiation control group were given physiological saline. The positive group was given 2 days before irradiation and once daily. The other groups were administered with 1 Times. One-time whole body irradiation with 60Co γ-rays was performed at a dose of 6 Gy at a dose rate of 0.8 Gy / min. On the 14th day after irradiation, the peripheral blood of mice was collected and the serum was separated to detect the activity of SOD and the content of MDA. Results Serum SOD activity in peripheral blood of BSH low, medium and high dose groups was significantly higher than that of irradiated control group (P <0.05), serum MDA level and low dose group were significantly lower than those of irradiated control group (P <0.05) High-dose group was significantly lower than the irradiated control group (P <0.01). Conclusion BSH has a significant anti-oxidant effect, suggesting that anti-oxidative capacity of BSH is one of the mechanisms of its anti-radiation effects.
