通过检测大鼠腹腔巨噬细胞(M)胞浆游离钙离子浓度([Ca2+]i)和45Ca内流与外排研究了不同浓度白芍总甙(TGP)对MCa2+跨膜转运的影响.结果表明,胞外有Ca2+时,TGP(2.5-20mgL-1)对亚适浓度卡西霉素(0.1μmolL-1),脂多糖(LPS,5mgL-1)和甲酰三肽(fMLP,1μmolL-1)升高M的[Ca2+]i具有明显的增强作用;而胞外无Ca2+时,TGP对LPS或fMLP刺激胞内钙池释放Ca2+引起[Ca2+]i升高的作用无明显影响;TGP可使卡西霉素刺激M的45Ca内流明显增加,同时较高浓度(10-40mgL-1)亦浓度依赖性促进45Ca外排.这些结果提示,TGP有促进胞外钙内流作用,较高浓度TGP可促进胞内钙外排,从而降低[Ca2+]i.
The effects of different concentrations of total glucosides of paeony on the migration of MCa2 + transmembrane were studied by measuring the intracellular free calcium concentration ([Ca2 +] i) and the 45Ca influx and efflux in rat peritoneal macrophages (M) influences. The results showed that TGP (2.5-20 mg L-1) had no effect on Ca2 +, Ca2 +, Ca2 +, Ca2 + The effect of TGP on [Ca2 +] i release from Ca2 + -induced intracellular Ca2 + -increased by LPS or fMLP in vitro without Ca2 + TGP significantly increased 45Ca Ca2 + influx in M and higher concentration (10-40mgL-1) promoted Ca2 + efflux in a dose-dependent manner. These results suggest that TGP promotes extracellular calcium influx and that higher concentrations of TGP promote intracellular calcium efflux and thereby reduce [Ca2 +] i.
