目的 :探讨rhIL 1β对人胚胎胰岛分泌IL 6以及对胰岛分泌功能的影响。 方法 :采用常规消化方法分离人胚胎胰岛并对其进行原代培养 ,加入不同浓度的rhIL 1β培养 48h取其上清 ,测定IL 6、胰岛素、胰高血糖素含量和IL 6McAb中和试验 ,对rhIL 1β刺激和对照组胰岛RNA进行IL 6RNA斑点杂交。 结果 :(1)经rhIL 1β刺激后胰岛培养上清IL 6含量显著升高 ,在一定范围内呈剂量依赖关系 (74~ 2 94mU/胰岛 ) ;(2 )IL 6McAb对上清中IL 6有很强的中和效应 ;(3)斑点杂交结果显示rhIL 1β刺激的胰岛RNA中IL 6mRNA含量升高 ;(4 )上清胰岛素和胞内胰岛素含量降低 (分别为 0 78~ 0 48IU/胰岛和 0 79~ 0 45IU/胰岛 ) ;(5 )上清胰高血糖素分泌能力明显升高 (1 0~ 3 2pg/胰岛 ) ,分泌趋势与IL 6相似。结论 :胰岛的分泌功能及产生IL 6的能力受IL 1的调节
AIM: To investigate the effect of rhIL 1β on the secretion of IL-6 from human embryonic islets and its function of islet secretion. Methods: Human embryonic islets were isolated by routine digestion method and cultured in primary culture. After different concentrations of rhIL 1β were added to culture for 48 hours, the supernatant was collected and the IL 6, insulin, glucagon and IL 6McAb neutralization were tested. rhIL 1 β stimulation and control islet RNA were subjected to IL 6 RNA dot blotting. Results: (1) The content of IL-6 in islet culture supernatant was significantly increased after stimulated by rhIL 1β, and in a dose-dependent manner (74-294mU / islet); (2) The effect of IL 6McAb on IL 6 in supernatant (3) The result of dot hybridization showed that the content of IL 6 mRNA in islet RNA stimulated by rhIL 1β increased; (4) the content of supernatant insulin and intracellular insulin decreased (0 78 ~ 0 48 IU / islet and 0 79 ~ 0 45IU / islet); (5) The secretion of glucagon in supernatant was significantly increased (10 ~ 32 pg / islet), the secretion tendency was similar to IL 6. CONCLUSIONS: The ability of islet secretion and IL 6 production is regulated by IL 1
