目的:研究中药温化蠲痹方对胶原诱导性关节炎(CIA)大鼠滑膜血管内皮生长因子(VEGF)表达的影响,探讨温化蠲痹方治疗CIA的作用机制。方法:选用健康雄性Wistar大鼠,随机分为造模组、正常对照组(10只,简称正常组),造模组采用牛Ⅱ型胶原乳剂于大鼠尾根部注射,建立CIA模型。再选取造模成功大鼠30只随机分为模型组(10只)、MTX组(10只)、温化蠲痹方组(10只,简称中药组)。中药组给予温化蠲痹方22.9 g/(kg·d)灌胃,模型组灌胃生理盐水,均每天1次,MTX组按0.78mg/kg剂量灌胃MTX混悬液,每周1次,连续30天。给药结束后,运用免疫组化法检测大鼠滑膜VEGF水平。结果:与模型组比较,治疗后中药组和MTX组大鼠关节炎指数(AI)评分明显降低(P<0.01);与治疗前比较,中药组和MTX组大鼠AI评分均明显降低(P<0.01)。与正常组比较,模型组大鼠滑膜VEGF表达水平明显升高(P<0.01);与模型组比较,中药组和MTX组大鼠滑膜VEGF表达水平明显降低(P<0.01),中药组和MTX组之间比较差异无统计学意义(P>0.05)。结论:中药温化蠲痹方可能通过下调CIA大鼠滑膜VEGF水平,抑制血管翳形成而治疗CIA。
Objective: To investigate the effect of Wenhua Huibi Recipe on the expression of vascular endothelial growth factor (VEGF) in synovial membrane of rats with collagen-induced arthritis (CIA) and to explore the mechanism of action of Wenhua Danbi recipe on CIA. Methods: Healthy male Wistar rats were randomly divided into model group and normal control group (n = 10, referred to as normal group). The model rats were injected with the tail of the rats with type Ⅱ collagen emulsion to establish CIA model. Thirty rats were selected and randomly divided into model group (n = 10), MTX group (n = 10) and Wenhuahubi group (n = 10). The rats in the traditional Chinese medicine group were treated with 22.5 g / (kg · d) of Wenhua Xiebi Decoction. The rats in the model group were fed with normal saline once a day. The MTX group was orally administered with MTX suspension at a dose of 0.78mg / kg once a week For 30 consecutive days. After administration, the level of VEGF in synovium of rats was detected by immunohistochemistry. Results: Compared with the model group, the arthritis index (AI) scores of the Chinese medicine group and the MTX group were significantly decreased after treatment (P <0.01). Compared with the pretreatment group, the AI scores of the Chinese medicine group and the MTX group were significantly decreased <0.01). Compared with normal group, the expression of VEGF in synovium of model group was significantly increased (P <0.01). Compared with model group, the expression of VEGF in synovium of Chinese medicine group and MTX group was significantly decreased (P <0.01) There was no significant difference between MTX group and MTX group (P> 0.05). CONCLUSION: Wenhua Pianbi prescription may treat CIA by down-regulating the level of VEGF in synovium of CIA rats and inhibiting angiogenesis.
