目的观察解毒祛瘀方联合化疗对EMT-6乳腺癌细胞移植瘤小鼠细胞株凋亡和COX-2表达的影响。方法 BALB/c小鼠接种乳腺癌细胞株EMT-6,造模成功后分为模型组、解毒祛瘀方组、化疗组、解毒祛瘀方加化疗组,每组10只,分别给予相应药物干预,共用药14天,停药后24h处死动物取材;计算抑瘤率,应用流式细胞术双标法检测其细胞凋亡率,应用免疫组化技术检测各干预组对EMT-6乳腺癌细胞中COX-2蛋白表达的影响。结果与解毒祛瘀方组比较,化疗组对乳腺癌小鼠抑瘤率较高,差异有统计学意义(P<0.05);与解毒祛瘀方组和化疗组比较,解毒祛瘀方加化疗组对乳腺癌小鼠抑瘤率最高,差异有统计学意义(P<0.05)。解毒祛瘀方组能诱导EMT-6乳腺癌细胞凋亡,作用较化疗组弱,而解毒祛瘀方加化疗组则能够进一步增强化疗药物对凋亡的影响。与模型组比较,各用药组COX-2表达均降低,差异有统计学意义(P<0.05,P<0.01);与解毒祛瘀方组、化疗组比较,解毒祛瘀方加化疗组COX-2的表达下调最明显,差异有统计学意义(P<0.05,P<0.01)。结论解毒祛瘀方联合化疗有一定抑瘤、促凋亡作用,其机制可能与下调乳腺癌细胞COX-2的表达有关。
Objective To observe the effect of Jiedu Quyu Decoction combined with chemotherapy on cell apoptosis and COX-2 expression in EMT-6 breast cancer xenografts in mice. Detoxification Quxufang group can induce EMT-6 breast cancer cell apoptosis, the effect is weaker than the chemotherapy group, and detoxification and stasis plus chemotherapy group can further enhance the impact of chemotherapy drugs on apoptosis. Compared with the model group, the COX-2 expression of each drug group decreased, the difference was statistically significant (P <0.05, P <0.01); compared with the Jiedu Qushi Decoction group and the chemotherapy group, 2 expression was the most significant, the difference was statistically significant (P <0.05, P <0.01). Conclusions Jiedu Quyu Decoction combined with chemotherapy has anti-tumor and pro-apoptotic effects, and its mechanism may be related to the down-regulation of COX-2 expression in breast cancer cells.
