目的观察吲哚乙酸联合辣根过氧化物酶对Tca8113细胞移植瘤的作用。方法建立Tca8113细胞裸鼠移植瘤模型,随机分为阴性对照组(瘤内注射无菌生理盐水)、阳性对照组(腹腔注射5-Fu,40mg/kg)和实验组(HRP:24mg/L,IAA:2mM,各0.1ml瘤内注射),2次/周,给药3周,计算肿瘤生长抑制率,免疫组化检测肿瘤中PCNA和Bcl-2基因表达情况,HE染色观察裸鼠脏器损害情况。结果阳性对照组和实验组的抑瘤率分别为56.02%、49.21%,与阴性组对照比较有统计学意义(P<0.05)。阳性对照组和实验组裸鼠肿瘤组织中的PCNA、Bcl-2的表达均低于阴性对照组,与之比较有明显差异(P<0.05);实验组裸鼠脏器未发现明显损害。结论吲哚乙酸联合辣根过氧化物酶能够抑制Tca8113移植瘤的生长,其机制可能是促进肿瘤细胞发生凋亡(对PCNA、Bcl-2表达的抑制);其短期内对Tca8113荷瘤裸鼠的体重要脏器无明显影响。
Objective To observe the effect of indoleacetic acid combined with horseradish peroxidase on Tca8113 cell xenografts. Methods Tca8113 cells in nude mice model were established and randomly divided into negative control group (intratumoral injection of sterile saline), positive control group (intraperitoneal injection of 5-Fu, 40mg / kg) and experimental group (HRP: 24mg / The tumor growth inhibition rate was calculated by immunohistochemistry.
