目的:观察四逆汤含药血清对阿霉素(ADR)引起的培养乳鼠心肌细胞毒性的影响。方法:采用原代培养的Wister大鼠乳鼠心肌细胞,以1.0μg/mL的ADR造成急性心肌细胞中毒模型,测定心肌细胞乳酸脱氢酶(LDH)漏出量、心肌细胞丙二醛(MDA)含量、心肌细胞搏动频率及MTT分析法测定心肌细胞活力,测定过氧化氢酶(CAT)、总超氧化物歧化酶(T-SOD)、超氧化物歧化酶(Cu-SOD),观察大鼠生存率的变化。结果:四逆汤含药血清对ADR引起的心肌细胞损伤可减少LDH漏出量和心肌细胞内MDA含量,并改善心肌细胞活力和搏动频率,改善Cu-SOD的活性,延长大鼠生存期。结论:四逆汤含药血清对ADR引起的心肌细胞损伤有一定的保护作用,其作用机制与其抑制ADR引起的脂质过氧化损伤有关。
Objective: To observe the effects of serum containing Sini Decoction on doxorubicin (ADR) -induced cardiomyocyte cytotoxicity in cultured neonatal rat cardiomyocytes. Methods: Primary cultured cardiomyocytes of Wister rats were used to induce acute cardiomyocyte poisoning with ADR at 1.0 μg / mL. Cardiac myocyte leakage of lactate dehydrogenase (LDH), malondialdehyde (MDA) The cardiomyocyte activity was measured by MTT assay. The levels of catalase (CAT), total superoxide dismutase (T-SOD) and superoxide dismutase (Cu-SOD) Survival rate changes. Results: Sini Decoction-containing serum on ADR-induced myocardial cell injury can reduce LDH leakage and myocardial MDA content and improve myocardial cell viability and pulsatile frequency, improve the activity of Cu-SOD and prolong the survival of rats. Conclusion: Sini Decoction-containing serum can protect cardiomyocytes against ADR to a certain extent, and its mechanism may be related to the inhibition of lipid peroxidation induced by ADR.
