目的:探讨整联蛋白亚基在喉鳞状上皮细胞癌(LSCC)中的表达情况及其意义。方法:采用基因芯片检测4例LSCC及对应癌旁正常组织中整联蛋白亚基的表达。采用RT-PCR检测24例LSCC及对应癌旁正常组织中基因芯片筛选出的4例均有明显差异表达的Integrins subunits。结果:基因芯片检测出IntegrinαV、Integrinβ8、Integrinα8,在LSCC组织及对应癌旁正常组织中均有明显差异表达,其中IntegrinαV、Integrinβ8表达上调,Integrinα8表达下调。RT-PCR检测IntegrinαV在LSCC中的相对表达量为1.0131±0.4780,与在对应癌旁正常组织中的相对表达量(0.7591±0.4678)比较差异有统计学意义(P<0.05);Integrinβ8在LSCC中的相对表达量为1.7362±1.3849,与在对应癌旁正常组织中的相对表达量(1.2267±0.9363)比较差异有统计学意义(P<0.05);Integrinα8在LSCC中的相对表达量为(0.2646±0.2622),与对应癌旁正常组织中的相对表达量(0.5457±0.3827)比较差异也有统计学意义(P<0.05)。结论:IntegrinαV、Integrinβ8及Integrinα8可能与LSCC的发生、发展相关。
Objective: To investigate the expression of integrin subunits in laryngeal squamous cell carcinoma (LSCC) and its significance. Methods: The gene chip was used to detect the expression of integrin subunits in 4 cases of LSCC and corresponding normal tissues. RT-PCR was used to detect the expression of Integrins subunits in 4 cases of LSCC and corresponding normal tissue adjacent to normal tissue. Results: IntegrinαV, Integrinβ8 and Integrinα8 were detected by microarray. The expression of IntegrinαV, Integrinβ8 and Integrinα8 were significantly down-regulated in LSCC tissues and adjacent normal tissues. The relative expression level of IntegrinαV in LSCC by RT-PCR was 1.0131 ± 0.4780, which was significantly different from that in the corresponding adjacent normal tissues (0.7591 ± 0.4678) (P <0.05). The expression of Integrinβ8 in LSCC (1.7362 ± 1.3849, P <0.05). The relative expression level of Integrinα8 in LSCC was (0.2646 ± 0.383) 0.2622), which was also significantly different from the corresponding normal tissue (0.5457 ± 0.3827) (P <0.05). Conclusion: IntegrinαV, Integrinβ8 and Integrinα8 may be related to the occurrence and development of LSCC.
