目的观察慢性心力衰竭大鼠肝脏、脂肪及骨骼肌的胰岛素受体(InsR)及其亚型,IRS-1、IRS-2,葡萄糖转运蛋白-1(GLUT-1)和GLUT-4 mRNA的表达及其变化趋势。方法建立腹主动脉缩窄诱导大鼠心力衰竭模型,行腹腔注射胰岛素耐量实验(IPITT),静脉葡萄糖耐量实验(WGTT)和静脉胰岛素释放实验(IRT)。RT-PCR检测各组组织InsR、IRS,GLUT mRNA的表达。结果与假手术组(SHAM)比较,心力衰竭组(CHF)IVGTT、IPITT、IRT结果异常。肝脏和骨骼肌中InsR[(1.02±0.09)vs(2.23±0.33);(0.003±0.000)vs(0.01±0.01),P<0.05]及脂肪中各基因mRNA含量均降低(P<0.05)。结论慢性心力衰竭可引起IR,胰岛素通路成分mRNA发生改变。
Objective To observe the expression of insulin receptor (IRR), IRS-1, IRS-2, GLUT-1 and GLUT-4 mRNA in liver, adipose and skeletal muscle of rats with chronic heart failure And its changing trend. Methods The rat models of heart failure induced by abdominal aorta constriction were established. IPITT, WGTT and IRT were performed. The mRNA expression of InsR, IRS and GLUT in each group was detected by RT-PCR. Results The results of IVGTT, IPITT and IRT in patients with heart failure (CHF) were abnormal compared with those in sham operation group (SHAM). In the liver and skeletal muscle, InsR [(1.02 ± 0.09) vs (2.23 ± 0.33); (0.003 ± 0.000) vs (0.01 ± 0.01), P <0.05] and adipose mRNA levels decreased (P <0.05). Conclusion Chronic heart failure can cause IR, insulin pathway component mRNA changes.
