目的:观察研究枸杞多糖(LyciumBarbarumPolysacharides,LBP)对37℃体外培养的小鼠睾丸组织形态结构有无保护作用。方法:未加LBP的培养组为对照组,添加500μg/mlLBP的培养组为实验组。昆明种小白鼠睾丸组织在32℃和37℃CO2培养箱中分别培养24、48和72h后制成石蜡切片,HE染色,光镜下观察分析。结果:32℃培养的小鼠睾丸组织对照组与实验组在各时间内曲精小管结构变化无显著差异。而37℃培养的小鼠睾丸组织曲精小管结构48h之内两组间亦无明显差异。但72h时,对照组显示曲精小管上皮结构紊乱严重,大量细胞脱落退化;而实验组曲精小管上皮结构变化与32℃培养72h的组织结构相似,生精上皮仍为复层,管腔脱落细胞明显少于对照组。结论:LBP可减弱由于培养温度过高所致的曲精小管结构损伤。
OBJECTIVE: To observe the protective effect of Lycium barbarum Polysacharides (LBP) on the morphological structure of mouse testis in vitro cultured at 37 °C. Methods: The culture group without LBP was used as the control group, and the culture group with 500 μg/ml LBP was added as the experimental group. The testes of Kunming mice were cultured for 24, 48, and 72 hours at 32°C and 37°C CO2 incubator, respectively, and paraffin sections were made. HE staining was performed under light microscope. RESULTS: There was no significant difference in the structure of the seminiferous tubules between the control group and the experimental group at 32 °C. There was no significant difference between the two groups within 48h of the testis tissue of the testis tissue cultured at 37°C. At 72 hours, the control group showed severe disruption of the structure of the epithelial cells of the seminiferous tubules, and a large number of cells were degenerated. However, the epithelial structure of the seminiferous tubules in the experimental group was similar to that of the tissue culture at 32°C for 72 hours. The seminiferous epithelium was still multilamellar, and the epithelial cells were exfoliated. Significantly less than the control group. Conclusion: LBP can attenuate the structural damage of the seminiferous tubules due to the high culture temperature.
