目的 :研究雷公藤多甙 (tripterygiumwilfordiihookfmulti glycosides ,TWG)对角朊细胞株 (COLO 16 )的增殖抑制及对凋亡的影响 ,并探讨其治疗免疫性皮肤病的机制。方法 :MTT法检测细胞增殖抑制 ,二苯胺及PI染色法检测细胞凋亡 ,RT PCR测细胞bcl 2及bcl XsmRNA表达。结果 :TWG 0 .2 5mg·L-1与角朊细胞共育 72h后 ,角朊细胞增殖活性受抑 ,此抑制作用具药物 (质量 )浓度依赖性 ;TWG 1mg·L-1作用于角朊细胞 72h ,即可使片段化DNA含量及亚二倍体细胞百分数较对照明显增加 ,此现象随药物质量浓度增加而增高 ;TWG 5mg·L-1,可上调角朊细胞bcl XsmRNA表达 ,此作用随药物作用时间延长而增强 ;TWG对bcl 2mRNA表达则无明显影响。结论 :TWG可抑制角朊细胞株增殖并诱导其凋亡 ,此诱导凋亡机制可能与bcl Xs上调有关
OBJECTIVE: To study the effects of tripterygium wilfordii hookfmulti glycosides (TWG) on the proliferation and apoptosis of keratinocyte cell lines (COLO 16) and to explore its mechanism for the treatment of autoimmune skin diseases. Methods: MTT assay was used to detect the cell proliferation inhibition. Apoptosis was detected by diphenylamine and PI staining. The expression of bcl 2 and bcl-2 mRNA was detected by RT PCR. Results: TWG 0. 25mg · L-1 and keratinocyte co-treatment 72h, keratinocyte proliferation activity was inhibited, the inhibition with drug (mass) concentration-dependent; TWG 1mg · L-1 role in keratin 72h, can make the fragment DNA content and sub-diploid cell percentage significantly increased compared with the control, this phenomenon increases with the increase of drug concentration; TWG 5mg · L-1, can raise keratinocyte bcl XsmRNA expression, this role TWG increased the expression of bcl-2 mRNA with the prolongation of drug effect. Conclusion: TWG can inhibit the proliferation and induce the apoptosis of keratinocyte cell lines, which may be related to the up-regulation of bcl Xs
