目的探讨高迁移率蛋白B1(HMGB1)、基质金属蛋白酶-9(MMP-9)m RNA和蛋白在子宫内膜癌组织中表达水平及临床意义。方法应用实时荧光定量RT-PCR及Western印记法检测正常子宫内膜组织、单纯型增生症子宫内膜组织、非典型增生子宫内膜组织和子宫内膜癌Ⅰ期、Ⅱ期及Ⅲ期组织中HMGB1、MMP-9 m RNA和蛋白的表达水平。结果与正常子宫内膜组织比较,子宫内膜癌组织HMGB1、MMP-9 m RNA及蛋白的表达量明显升高,差异有统计学意义(P<0.05);子宫内膜癌组织HMGB1、MMP-9蛋白的表达上调与手术病理分期、淋巴结转移及浸润深度有关(P<0.05),与癌组织的分化程度无关(P>0.05);子宫内膜癌组织中,HMGB1蛋白与MMP-9蛋白的表达呈正相关(r=0.900,P<0.05)。结论 HMGB1、MMP-9在子宫内膜癌的发生、发展及转移的过程中起着重要的作用,其有望成为子宫内膜癌的早期诊断及预后评价的分子标志物。
Objective To investigate the expression of HMGB1, MMP-9 mRNA and protein in endometrial carcinoma and its clinical significance. Methods Real-time fluorescent quantitative RT-PCR and Western blotting were used to detect the expressions of MMP-2 in normal endometrium, simple hyperplastic endometrium, atypical hyperplasia endometrial tissue and endometrial carcinoma in stages Ⅰ, Ⅱ and Ⅲ HMGB1, MMP-9 m RNA and protein expression levels. Results Compared with normal endometrial tissue, the expression of HMGB1, MMP-9 mRNA and protein in endometrial carcinoma tissues was significantly increased (P <0.05). The expressions of HMGB1, MMP- (P <0.05), but not with the degree of differentiation (P> 0.05). The expression of HMGB1 and MMP-9 in endometrial carcinoma The expression was positively correlated (r = 0.900, P <0.05). Conclusions HMGB1 and MMP-9 play an important role in the development, progression and metastasis of endometrial carcinoma, which are expected to be the molecular markers for the early diagnosis and prognosis evaluation of endometrial cancer.
