目的:探讨骨髓增生异常综合征(MDS)患者骨髓中HOXA9基因和蛋白的表达模式及其与诊断、治疗的关系。方法:应用流式细胞术检测33例MDS患者、12例初治急性髓系白血病、20例免疫性血小板减少性紫癜和18名正常对照者的骨髓样本的HOXA9蛋白水平,利用实时荧光定量PCR检测HOXA9 mRNA的表达水平。结果:MDS患者HOXA9~+/CD34~+和HOXA9~+/CD34~+CD38~-比例均高于对照组(P<0.05)。HOXA9的mRNA相对表达量与蛋白水平相一致。未行地西他滨治疗的患者HOXA9~+/CD34~+为(50.64±27.59)%,HOXA9~+/CD34~+CD38~-为(55.67±28.57)%,均高于正常对照组(P<0.05)。经地西他滨治疗患者的HOXA9~+/CD34~+和HOXA9~+/CD34~+CD38~-比例均低于未治组(P<0.05)。结论:HOXA9在MDS中异常高表达且与相关临床特征有相关,提示HOXA9在MDS诊治中具有指导意义。
Objective: To investigate the expression pattern of HOXA9 gene and protein in bone marrow of patients with myelodysplastic syndrome (MDS) and its relationship with diagnosis and treatment. Methods: Flow cytometry was used to detect HOXA9 protein levels in bone marrow of 33 cases of MDS patients, 12 cases of newly diagnosed acute myeloid leukemia, 20 cases of immune thrombocytopenic purpura and 18 normal controls. Real-time fluorescent quantitative PCR HOXA9 mRNA expression levels. Results: The proportions of HOXA9 ~ + / CD34 ~ + and HOXA9 ~ + / CD34 ~ + CD38 ~ - in MDS patients were higher than those in control group (P <0.05). HOXA9 mRNA relative expression level consistent with the protein level. (50.64 ± 27.59)% and HOXA9 ~ + / CD34 ~ + CD38 ~ - (55.67 ± 28.57)%, respectively, in patients who were not treated with citalopram were higher than those in the normal control group <0.05). The rates of HOXA9 ~ + / CD34 ~ + and HOXA9 ~ + / CD34 ~ + CD38 ~ - in decitabine were lower than those in untreated group (P <0.05). CONCLUSION: HOXA9 is highly expressed in MDS and correlates with related clinical features, suggesting that HOXA9 may be instructive in the diagnosis and treatment of MDS.
