缺血性神经元损伤的性质 (凋亡或坏死 )是神经研究领域关注的热点。但目前用于区分凋亡与坏死细胞的TUNEL 技术 ,较易受主观因素的影响。本实验用图像分析技术对缺血动物脑的 TU NEL 染色切片进行多视场多参数形态定量分析 ,即细胞形态定量参数 (核面积、周长、直径、形状因子 )和含量光密度 (平均光密度、积分光密度 )。本实验结果显示图像分析技术能提高 TU NEL 鉴别凋亡与坏死细胞的灵敏度和精确性 ,并提供一种简便快捷的形态定量方法 ,值得在神经研究领域中推广应用。
The nature of ischemic neuronal damage (apoptosis or necrosis) is a hot area of focus in neuroscience research. However, the TUNEL technique currently used to differentiate apoptotic and necrotic cells is more susceptible to subjective factors. In this experiment, multi-field multi-parameter morphometric analysis of TU NEL-stained sections of ischemic brain was performed by using image analysis technique. The results showed that quantitative parameters of cell morphology (nuclear area, perimeter, diameter and shape factor) and content optical density Density, integral optical density). The experimental results show that the image analysis technology can improve the sensitivity and accuracy of TU NEL in identifying apoptotic and necrotic cells, and provide a simple and rapid method for morphometric quantification, which is worthy of promotion and application in the field of neurological research.
