目的:通过体外培养的方式,观察长效β2鄄受体激动剂福莫妥罗(formoterol)、白三烯受体拮抗剂孟鲁斯特(montelukast)和扎鲁斯特(zafirlukast)3种药物对哮喘患者外周血单个核细胞(PMBC)分泌白介素鄄4(IL鄄4)、白介素鄄5(IL鄄5)的影响。方法:哮喘患者26例,正常对照组10名。分离受试对象之外周血单个核细胞并接种于细胞培养板,药物孔相应加入montelukast、zafirlukast或formoterol预培养2h后,所有培养孔加植物血凝素(PHA)刺激培养48h,收集上清液,用ELISA法测定培养上清液中IL鄄4、IL鄄5的浓度并进行相应的统计学处理。结果:①哮喘组PBMC单纯经PHA刺激后,平均IL鄄4、IL鄄5的分泌水平高于健康组。②Montelukast、zafirlukast可导致哮喘组IL鄄5分泌下降,且有统计学意义。结论:Montelukast和zafirlukast均能抑制IL-5的分泌,前者还可能对IL鄄4的分泌也具有抑制作用,因而有利于纠正哮喘患者Th1/Th2的不平衡。本研究未见formoterol对IL鄄4、IL鄄5的分泌有影响。
OBJECTIVE: To observe the effects of long-acting β2-receptor agonist formoterol, leukotriene receptor antagonists montelukast and zafirlukast by in vitro culture On interleukin-4 (IL-4) and interleukin-5 (IL-5) secretion in peripheral blood mononuclear cells (PMBC) of asthmatic patients. Methods: 26 asthmatic patients and 10 normal controls. Peripheral blood mononuclear cells (PBMCs) were isolated and seeded on the cell culture plates. After pretreatment with montelukast, zafirlukast or formoterol for 2h, all the wells were incubated with phytohemagglutinin (PHA) for 48h. The supernatants The concentrations of IL-4 and IL-5 in the culture supernatant were measured by ELISA and the corresponding statistics were performed. Results: ① The PBMC of asthmatic group were stimulated by PHA only, the average level of IL-4 and IL-5 secretion was higher than that of healthy group. ② Montelukast, zafirlukast can lead to IL-5 secretion in asthma group decreased, and have statistical significance. Conclusion: Both Montelukast and zafirlukast can inhibit the secretion of IL-5, and the former may also inhibit the secretion of IL-4, which is helpful to correct the imbalance of Th1 / Th2 in asthmatic patients. This study did not find formoterol on IL Juan 4, IL Juan 5 secretion.
