目的研究特异性细胞毒T淋巴细胞(cytotoxicTlymphocyte,CTL)杀伤胃癌细胞BGC823的超微结构变化。方法用22例患者的胃腺癌细胞反复冻融后的滤液,抗CD3单克隆抗体、白细胞介素2(IL2)与患者自体外周血淋巴细胞共同培养,体外诱导CTL,然后将CTL与BGC823共同孵育,用电子显微镜观察CTL作用于BGC823后的超微结构改变。结果电镜观察可见CTL包围并破坏靶细胞,靶细胞凋亡的形态表现为异染色质浓缩呈特征性半月形帽状核,还可见靶细胞溶解坏死。结论特异性CTL可使靶细胞凋亡,溶解而导致其死亡
Objective To study the ultrastructural changes of specific cytotoxic T lymphocytes (CTL) in killing gastric cancer cells BGC823. Methods Twenty-two patients with gastric adenocarcinoma cells were repeatedly frozen and thawed. Anti-CD3 monoclonal antibody, interleukin-2 (IL-2) was co-cultured with autologous peripheral blood lymphocytes, and CTLs were induced in vitro. The BGC-823 was incubated together and electron microscopy was used to observe the ultrastructural changes after the CTLs acted on BGC-823. RESULTS: The electron microscopy showed that CTLs surrounded and destroyed the target cells. The morphology of the target cells showed a heterogeneous half-moon cap-like nucleus with heterochromatin concentration, and the target cells also showed lysis and necrosis. Conclusion The specific CTL can cause apoptosis in target cells, which leads to its death.
